Research overview

FOXO4-DRI is a synthetic peptide made of D-amino acids in reversed order, a D-retro-inverso design that resists enzymatic breakdown while mimicking the shape of part of the FOXO4 protein. It was designed to disrupt the interaction between FOXO4 and p53 that helps senescent cells survive.

In a 2017 study in Cell, the peptide selectively triggered apoptosis in senescent cells and, in aged mice, improved fitness, fur density and kidney function. That made it a widely cited tool in senolytic research, the study of compounds that clear senescent cells. Replication and selectivity across cell types are active research questions. Supplied lyophilized.
