Melanotan I, or afamelanotide, is a linear 13-amino-acid analogue of alpha-MSH with two substitutions, norleucine at position 4 and D-phenylalanine at position 7, that make it much more stable and potent than the natural hormone. It acts selectively at the MC1 receptor on melanocytes.

Afamelanotide is approved in the EU and the US as an implant for erythropoietic protoporphyria, a rare condition of severe light sensitivity, where raising eumelanin in the skin improves light tolerance. In research it is the reference MC1-selective agonist for studying melanogenesis and photoprotection. Supplied lyophilized; protect it from light.
